locked nucleic acid.锁核酸 (lockednucleicacid ,LNA)是一种新型的寡核酸衍生物 ,结构中 β D 呋喃核糖的 2’ O ,4’ C位通过缩水作用形成环形的氧亚甲基桥、硫亚甲基桥或胺亚甲基桥 ,呋喃糖的结构锁定在C3’内型的N构型 ,形成了刚性的缩合结构.LNA作为一种新的反义核酸 ,具有与DNA/RNA强大的杂交亲和力、反义活性、抗核酸酶能力、水溶性好及体内无毒性等优点.LNA在基因诊断和基因治疗上有很多优势 ,如 :单链核酸的多态性基因分型、LNA寡聚体具有高效抑制端粒酶活性及LNA修饰的DNA核酶 (LNAzymes)高效清除高级结构的RNA等 ,有良好的应用研究前景

锁定核苷酸（LNA）单体是一种与加至核糖体环上的外2´-O,4´-C-次甲基桥相似的构形限制的核苷酸。当与互补DAN和RNA杂交时，包含LNA单体的寡聚核苷酸显示出大大提高了的热稳定性，并被认为是在核酸双链上辨别出某一给定的混合序列的最有希望的候选者，也被认为是一种反义分子。在这里我们利用25°C时的停流吸收测量和融合曲线研究了一系列的DNA–DNA 和DNA–LNA八聚体的寡聚核酸双链的形成的动力学和热力学。我们研究了DNA 八聚体的 5´-CAGGAGCA-3´与其互补的DNA 八聚体的5´-TGCTCCTG-3´，以及与LNA八聚体的5´-TLGCTCCTG-3´ (LNA-1), 5´-TLGCTLCCTG-3´ (LNA-2) 和 5´-TLGCTLCCTLG-3´(LNA-3)[分别包含1,2，或3个胸腺嘧啶2´-O,4´-C-次甲基-( -呋喃核糖基)核酸单体，指定为TL]的反应。在所有的例子中均可见以kobs = 2×107M-1·s-1进行的快速二级相关反应。在25°C时，从融合曲线得到的双链的解离常数为：DNA–DNA, 10nM; DNA–LNA-1, 20nM; DNA–LNA-2, 2nM; DNA–LNA-3, 0.3nM;因此证明了由LNA诱导的大大提高了双链的稳定性。既然相关等级是全部相等的，此稳定性的提高归因于较低的复合物解离率。
